ALDEFLUOR™--用于检测和分离具有活性的干/祖细胞

ALDEFLUOR™ 简单、快速,用以检测具有活性的正常 或肿瘤干/祖细胞:

ALDEFLUOR™荧光试剂系统提供了一种不同于一般依靠细胞表型、 基于细胞内乙醛脱氢酶( ALDH)活性对干/祖细胞进行鉴定、评估 和分离的方法。含有高表达 ALDH的细胞具有干/祖细胞的特性,这些细胞包括CD34+细胞、CD133+细胞、Kit+细胞、谱系抗原阴性 (Lin-)细胞、集落形成细胞、长期培养起始细胞以及NOD/SCID亚群细 胞。ALDEFLUOR™专为人血样品优化,目前也被应用于非造血谱系 以及多个物种的干/祖细胞鉴定。

• 检测源自多种谱系细胞的正常/恶性前体细胞,包括造血、乳腺、头颈部、肺、肝、胰腺、子宫颈、卵巢、膀胱、前列腺、结肠和甲状腺等

ALDEFLUOR™的工作原理:

• 不带电荷的ALDH-底物,BAAA(BODIPY®-氨基乙醛)通过被动 扩散进入活细胞。
• BAAA被细胞内的ALDH转化为带负电荷的反应产物BAA(BODIPY®-甘氨酸),该产物滞留在细胞内,使细胞高表达 ALDH,呈现明亮的荧光。
• 高表达ALDH(ALDHbr)的细胞带有明亮的荧光,在流式细胞仪的 绿色荧光通道(520-540 nm)被检出。
• 由于只有具有完整细胞膜的细胞才可以保留ALDEFLUOR™的反应 产物,因此 ALDEFLUOR™只鉴定具有活性的ALDHbr细胞。
